I'd start by looking at all the services, not just Exchange, to see if
anything's crashed or otherwise not running. Look in Task Manager to see if
anything is causing high CPU usage or using an unusually high amount of
memory. Check your Exchange queues and badmail folder to see if anything
unusual is going on there. See if the server is otherwise responsive and
"acting normally" when you log in locally. And most importantly, check your
AV software in case some unusual activity related to virus scanning is
causing this.
You could also try shutting down and restarting your Exchange services.
(Personally, I'd try stopping the AV for a few mintues first to see if that
helps. I'm troubleshooting a problem where my server becomes unresponsive,
and cycling the AV fixes it immediately).
