Dont know where to start, I'm banging my head with this one.

Regular domain users without any special permissions get a 401.3 error
referencing "Access denined to to an ACL setting on the requested
resource"

When I try to browse it from my account (I have domain admin
priveleges) I can see the site no problem.

Where would I begin to try and troubleshoot this problem?

This is just a simple "public- all users" team site with no special
permissions or rules defined.

What happens when they try to access the site? Are they preseneted with a
login box? or do they get the error message directly? Do they get this
message after their login fails? If your users are trying to log in using
their Windows usernames, ask them to use their email address instead and see
if it works for them. Also, site should have site group "READER" added in
the permissions list otherwise no one except the admin will have access to
the site. Even if it's a public - all users site, you will have to add users
as READERS in the site permissions list.
