In Exch2000, we limited our SMTP connection access by only allowing specific
IP addresses under SMTP server properties>Access Tab>Connection>"Only the
list below".

In Exch2007 SP1, could we accomplish the same by entering the IP addresses
in the Receive Connectors of the Hub Transport under Server Configuration
(Client & Default)? FYI, we do not use the Edge Transport Server role.

Yes - Receive Connectors -> Network tab -> Remote IP address(es).
Or using the shell:
Set-ReceiveConnector "Connector Name" -RemoteIPRanges
"1.2.3.4","5.6.7.8","200.1.1.1-200.1.1.20"

Do we need to keep the entry for
0000:0000:0000:0000:0000:0000:0.0.0.0-ffff:ffff:ffff:ffff:ffff:ffff:255.255.255.255
or did we just allow all IP addresses again?

When we entered specific IP addresses and removed this, we stopped getting
inbound emails so we had to add this entry back.

That's for IPV6. For now it's okay to leave it as is.
